Effects of biogas slurry soaking seeds on seed germination and physiological characteristics of Astragalus membranaceus var.mongholicus seedling

    摘要:

    通过培养皿发芽试验,设计 5个沼液浓度水平(0%,10%,25%,50%,75%)和 3个浸种时间水平(4,5,6 h),探讨沼液浓度和浸种时间对蒙古黄芪种子发芽势、发芽指数、活力指数、根长、苗高、根重、叶绿素(Chl)含量、丙二醛(MDA)含量、抗氧化酶活性(CAT、POD、SOD)及根系活力的影响,以寻求最佳沼液浸种浓度和时间组合,为黄芪产量和质量的提高提供理论依据。结果表明,25%沼液浸种 5 h黄芪种子发芽势最高,显著高于清水浸种4、5 h处理( P<0.05);25%沼液浸种 5 h的发芽指数、活力指数、根长、根重,Chla、Chlb、Chla+Chlb含量,根系活力、POD活性、SOD活性均显著高于其他 14个浸种处理( P<0.05);25%沼液浸种4、6 h苗高均显著高于其余 13个处理( P<0.05);丙二醛含量与 CAT活性测定结果一致,均以 25%沼液浸种 5 h最低( P<0.05)。说明适宜的沼液浸种有利于黄芪种子萌发和幼苗生长。其中 25%沼液浸种 5 h对黄芪种子萌发和幼苗生长效果最佳。

    Abstract:

    The influences of different biogas slurry concentrations and soaking time on the seed germination and physiological characteristics of Astragalus membranaceusvar. mongholicus seedling were studied by the culture dish germinationexperiment.And germination power,germination index,vigor index,root and seedling length,root height,content ofchlorophyll and MDA,antioxidant enzyme activities and root activity in various biogas slurry concentrations(0%,10%,25%,50%,75%)and soaking time(4,5,6h)were recorded and analyzed with PCA analysis,and the best biogas slurry soaking concentration and time were explored to improve the yield and quality of Astragalus membranaceusvar. mongholicus.Theresults showed that,the germination power of seeds was the highest in 25% biogas slurry soaking for 5h,which was significantlyhigher than that in water soaking for 4 and 5h( P<0.05).The germinationindex,vigorindex,rootlength,rootweight,Chlacontent,Chlbcontent,Chla+Chlbcontent,rootactivity,POD and SOD activity of seeds under 25% biogas slurry soaked for5 h were significantly higher than the other 14 soakingtreatments( P<0.05).The seedling height of seeds under 25% biogasslurry soaked for 4 and 6 h was thehighest( P<0.05).MDA content and CAT activity of seeds under 25% biogas slurry soakedfor 5 h were thelowest( P<0.05).It can be concluded that appropriate biogas slurry soaking can improve the germination and growth of Astragalus membranaceusvar.mongholicus.As awhole,soaking for 5 h in 25% biogas slurry had the best effect on seed germination and growth of Astragalus membranaceusvar.mongholicusseedling.
