温室黄瓜连作土壤真菌多样性变化分析

Analysis of fungal community structure and function changes in soil of cucumber continuous cropping in greenhouse

    摘要:

    以温室黄瓜连作1、3、5和7年的根际土壤为研究对象,利用Illumina MiSeq高通量测序技术分析土壤真菌ITS1区域土壤微生物群落结构和多样性的变化,利用FUNGuild对菌群功能进行预测,同时测定不同连作年限土壤的理化性质和相关酶活性变化。结果表明:不同连作年限土壤真菌在门水平上的群落组成变化明显,子囊菌门在连作5和7年样品中相对丰度分别下降9.94%和28.24%,而在连作3年样品中无明显变化,担子菌门相对丰度在连作3年样品中显著降低后又逐年上升。随着连作年限的增加,LEfSe分析显示,土壤中起主要作用的真菌属水平变化明显,群落丰度先增后减,但群落多样性提升。对不同连作年限真菌群落进行功能预测,479个OUT被划分为8个真菌功能类群,约占总OTU数量的39.75%,腐生菌是主要的功能菌群,其次是病原菌/腐生/共生过渡型和植物病原菌等,共生菌和腐生菌的相对丰度在连作7年样品中最高。环境因子的聚类分析显示,碱解氮(AN)和脲酶(SUR)聚为一类,有效磷(AP)和速效钾(AK)聚为一类,土壤真菌群落与pH、AN、SUR和AP具有显著相关性(P<0.05),对真菌菌群影响表现为SUR>AP>AN>pH。

    Abstract:

    The rhizosphere soil of cucumber continuous cropping in greenhouse for 1,3,5 and 7 years was taken as the research object.The changes in the microbial community structure and diversity in the ITS1 region of soil fungi were analyzed by Illumina MiSeq high-throughput sequencing technology,and the function of the fungi was predicted by FUNGuild,at the same time,the changes of physicochemical properties and related enzyme activities in different continuous cropping years were determined.The results showed that the community composition of soil fungi at the phylum level changed significantly in different years of continuous cropping.The relative abundance of Ascomycota phylum did not change significantly in the third year samples but decreased by 9.94% and 28.24% in the fifth and seventh year samples.The relative abundance of Basidiomycota phylum decreased significantly and then increased year by year.The LEfSe analysis showed that the abundance of fungi in the soil increased and then decreased,but the community diversity increased.The functional prediction of the fungal communities in different years of continuous cropping showed that 479 OUTs were classified into 8 fungal functional groups,accounting for 39.75% of the total OTUs. Saprophytic bacteria are the main functional flora, followed by pathogenic bacteria/saprophytic/symbiotic transition type and plant pathogenic bacteria. The relative abundance of symbiotic bacteria and saprophytic bacteria is the highest in the seventh year continuous cropping samples. Clustering analysis of environmental factors showed that available nitrogen(AN)and urease(SUR)clustered into one group,available phosphorus(AP)and available potassium (AK)clustered into one group,and the soil fungal communities were significantly correlated with pH,AN,SUR and AP,with the effect on fungal flora as follows:SUR > AP > AN > pH.
