Screening of Vicia Villosa varieties with low phosphorus tolerance at seedling stage and evaluation of their phosphorus efficiency types

    摘要:

    筛选毛叶苕子耐低磷品种,并评价其磷效率类型,为毛叶苕子磷高效品种的选育和生产应用提供理论依据。采用苗期水培的方法,以22个毛叶苕子品种为供试材料,在低磷(2 μmol/L Pi)和正常磷(200 μmol/L Pi)处理下,测定毛叶苕子的主根长、株高、地上部干重、根系干重、根冠比、地上部全磷含量和磷累积量,通过主成分分析和聚类分析评价各毛叶苕子品种的耐低磷能力,并采用隶属函数法计算各毛叶苕子品种的磷效率综合值以评价其磷效率类型。采用盆栽试验进一步验证水培试验的筛选结果,挑选耐低磷及低磷敏感品种各2个,在低磷(0 mg/kg P2O5)和正常磷(200 mg/kg P2O5)处理下,测定株高、地上部干重和全磷含量,并计算磷利用率。在低磷及正常供磷的水培试验中,主根长、株高、地上部干重、根系干重、根冠比、地上部全磷含量和磷累积量在不同毛叶苕子品种间存在显著差异,其中地上部干重和磷累积量的变异系数最大。基于上述2个指标的聚类分析,将22个毛叶苕子品种分为耐低磷型、中间型和低磷敏感型3类,其中耐低磷型品种包括徐苕3号、法国毛苕、78-171毛苕、山东早熟苕、沪3-1毛苕和78-103毛苕,低磷敏感型品种包括81-245毛苕、青苕1号、苏联苕子和74-60毛苕。基于磷效率综合值,将22个毛叶苕子品种的磷效率类型划分为正常磷高效低磷高效型(双高效型)、正常磷低效低磷低效型(双低效型)、正常磷高效低磷低效型和正常磷低效低磷高效型4类,其中双高效型品种有徐苕3号、法国毛苕、徐苕2号、沪3-1毛苕、特早熟毛苕和山东早熟苕,双低效型品种有葡萄牙毛苕、苏联苕子、81-243毛苕、新疆毛苕、青苕1号、内毛叶苕、关中毛苕、81-84毛苕和81-245毛苕。盆栽试验中,在正常磷处理下,徐苕3号和法国毛苕的株高、地上部干重和磷累积量都显著高于青苕1号及81-245毛苕;在低磷胁迫下,徐苕3号和法国毛苕的株高和地上部磷利用效率均显著高于青苕1号和81-245毛苕。综上,地上部干重和磷累积量可作为毛叶苕子苗期耐低磷性的评价指标。徐苕3号、法国毛苕、沪3-1毛苕和山东早熟苕属于耐低磷且磷高效型品种;81-245毛苕、青苕1号和苏联苕子属于低磷敏感且磷低效型品种。

    Abstract:

    In order to provide a theoretical basis for the breeding of high P-efficiecy varieties and the research on the physiological mechanism of plant tolerance to low phosphorus,Vicia Villosa varieties with low phosphorus tolerance were screened and their phosphorus efficiency types were evaluated.22 Vicia villosa varieties were tested through nutrient solution culture method,main root length,plant height,shoot dry weight,root dry weight,root-shoot ratio,shoot total phosphorus content and phosphorus accumulation were measured under the treatments of low phosphorus(2 μmol/LPi)and normal phosphorus(200 μmol/L Pi).The principal component analysis and cluster analysis were used to identify the low phosphorus tolerance ability of different Vicia Villosa cultivars,and the comprehensive value of phosphorus efficiency was calculated by the membership function method to evaluate the types of phosphorus efficiency.Pot experiment was conducted to further verify the results of nutrient solution culture test.Two cultivars with low phosphorus tolerance and two low phosphorus sensitive cultivars were selected as test materials,respectively.Plant height,shoot dry weight,shoot phosphorus content and phosphorus use efficiency were measured under low phosphorus(0 mg/kg P2O5)and normal phosphorus(200 mg/kg P2O5)treatments.Under low phosphorus or normal phosphorus treatment,there was a significant difference in main root length,plant height,shoot dry weight,root dry weight,root-shoot ratio,shoot total phosphorus content and phosphorus accumulation among different varieties,and the variation coefficient of shoot dry weight and phosphorus accumulation at the seedling period was the largest.Through cluster analysis of shoot dry weight and phosphorus accumulation,22 varieties of Vicia Villosa were divided into three types:low phosphorus tolerance type,intermediate type and low phosphorus sensitive type.The varieties with low phosphorus tolerance type included Xu Shao 3,French,78-171 Vicia Villosa,Shandong early maturing,Hu 3-1 and 78-103 Vicia Villosa.The varieties with low phosphorus sensitive type include 81-245 Vicia Villosa,Qing Shao 1,Soviet and 74-60 Vicia Villosa.According to the analysis of comprehensive value of phosphorus efficiency,22 cultivars of Vicia Villosa were divided into four types:high P-efficiency type under both low-Pi and normal-Pi treatments(double high P-efficiency type),low P-efficiency type under both low-Pi and normal-Pi treatments(double low P-efficiency type),low P-efficiency under low-Pi stress and high P-efficiency under normal-Pitreatment type and high P-efficiency under low-Pi stress and low P-efficiency under normal-Pi treatment type.The double high P-efficiency type included Xu Shao 3,French,Xu Shao 2,Hu 3-1,Super early maturing varieties and Shandong early maturing Vicia Villosa.Double low P-efficiency type included Portugal,Soviet,81-243 Vicia Villosa,Xinjiang,Qing Shao1,Inner Mongolia,Guanzhong,81-84 and 81-245.In pot experiment,under normal phosphorus treatment,the plant height,shoot dry weight and phosphorus accumulation of Xu Shao 3 and French Vicia Villosa were significantly higher than that of Qing Shao 1 and 81-245 Vicia Villosa; Under low phosphorus treatment,the plant height and phosphorus utilization efficiency of Xu Shao 3 and French Vicia Villosa were significantly higher than that of Qing Shao 1 and 81-245 Vicia Villosa.The shoot dry weight and phosphorus accumulation might be used as screening indexes for Vicia Villosa varieties with low phosphorus tolerance.Xu Shao 3,French,Hu 3-1 and Shandong early maturing Vicia Villosa were classified into low phosphorus tolerance and high phosphorus efficiency varieties.81-245,Qing Shao 1 and Soviet Vicia Villosa were classified into low phosphorus sensitive and low phosphorus efficiency varieties.
