Isolation,identification and growth characteristics of garlic root rot pathogen Alternaria embellisia DS55-6F

    摘要:

    大蒜是药食两用的百合科葱属作物,虽然具有抗肿瘤、抑菌等作用,但是连续种植后,常出现根部腐烂的现象。从山东省济宁市金乡县大蒜腐烂根样品中分离获得菌株DS55-6F,通过ITS、EF-1和gpd多基因分子生物学鉴定和形态学鉴定,明确该菌株为艾氏链格孢Alternaria embellisia;温室条件下,将该菌株回接大蒜幼苗,40 d后大蒜发生严重根部腐烂,通过重新分离和鉴定获得DS55-6F,符合柯赫氏验证试验,说明菌株艾氏链格孢Alternaria embellisia DS55-6F是大蒜根腐病病原菌。通过不同温度、不同pH、不同碳源、不同氮源4项指标对其生长特性进行评价,发现该菌株在28℃、pH 6、唯一碳源为麦芽糖、唯一氮源为牛肉浸粉或硝酸钾时菌丝生长较好,唯一碳源为乳糖、唯一氮源为硫酸铵或氯化铵时菌丝生长缓慢。

    Abstract:

    Garlic is a food and medicine Allium crop of Liliaceae.Although it has anti-tumor and bacteriostatic effects,root rot often occurs after continuous planting.The strain DS55-6F was isolated from garlic rotten root samples in Jinxiang county,Jining city,Shandong province.Through ITS,EF-1 and gpd gene molecularl identification and morphological identification,it was confirmed that the strain DS55-6F was Alternaria embellisia.Based on greenhouse experiments,the strain was inoculated to garlic seedlings,and serious root rot occurred in garlic after 40 days.DS55-6F was obtained through re-isolation and identification,which was in line with Koch’s verification test,indicating that Alternaria embellisia DS55-6F was the pathogen of garlic root rot.The growth characteristics were evaluated by four indexes:different temperature,different pH,different carbon sources and different nitrogen sources.The results showed that the strain was most conducive to mycelial growth at 28 ℃,pH 6,the only carbon source was maltose and the only nitrogen source was beef powder,and when the only carbon source was lactose,and the only nitrogen source was ammonium sulfate or ammonium chloride,the mycella grew slowly.
