    摘要:

    为研究不同木霉菌对栽培人参土壤的改良作用,分析木霉菌对土壤养分和土壤真菌群落结构与功能特征的影响。采用人参盆栽试验,添加外源木霉菌(钩状木霉DH3、MM3、MM4、Q7 4种菌株和哈茨木霉MM6菌种)后,分析土壤理化性质以及土壤微生物群落多样性变化。添加不同外源木霉菌处理后人参土壤理化性质发生变化,土壤pH值无显著变化,土壤全氮、有机质、铵态氮、硝态氮、有效磷、速效钾含量显著增加(P<0.05),而碳氮比(C/N)在添加木霉菌处理中降低,其中钩状木霉MM3和哈茨木霉MM6两株菌的理化性质表现最好。ITS测序分析获得703个OTUs,涵盖了8门、26纲、46目、86科、141属。木霉菌处理后土壤真菌微生物α多样性指数(ACE、Chao1)增加,尤其是钩状木霉MM3;钩状木霉MM3中的主要特征种群是肉座菌科(Hypocreaceae)和木霉菌属(Trichoderma),哈茨木霉MM6的主要特征种群是毛壳菌科(Chaetomiaceae)。冗余分析(RDA)发现,土壤 C/N 和硝态氮是影响土壤环境中微生物多样性的主要因素;相关分析表明,刺球菌属(Chaetosphaeria)与土壤全氮含量呈显著正相关,与C/N呈显著负相关。本结果为栽培人参土壤微生物改良和生物防治提供理论依据。

    Abstract:

    Exploring Trichoderma like biological fungal fertilizer in soil improvement has become a research focus. In order to study the improvement effect of different Trichoderma spp. on soil of ginseng cultivation,the soil physicochemical properties,and soil fungal community structure and function were analyzed. Ginseng pot experiment was conducted to analyze soil physical and chemical properties and soil microbial community diversity in which exogenous Trichoderma strains (four strains of T. hamatum DH3,MM3,MM4,Q7 and one strain of T. harzianum MM6) were added. The results showed that the soil physicochemical properties of Panax ginseng were changed by different exogenous Trichoderma strains. The soil pH did not change significantly,and the total nitrogen,organic matter,ammonium nitrogen,nitrate nitrogen,available phosphorus and available potassium were increased significantly(P< 0.05),while soil carbon nitrogen(C/N) ratio was decreased in the treatments with addition of Trichoderma strains. Generally speaking,the soil physicochemical properties treated by T. hamatum MM3 and T. harzianum MM6 were the best. Using Illumina NovaSeq high-throughput sequencing platform,the sequence of ITS variation region of soil fungi was determined,and 703 OTUs were obtained from all samples.Fungal species detected in these samples covered 8 phyla,26 classes,46 orders,86 families and 141 genera.The soil fungi α diversity index (ACE,Chao1) increased,especially in the treatment of T. hamatum MM3. The dominant fungal community were Hypocreaceae and Trichoderma in the treatment of T. hamatum MM3,and were Chaetomiaceae in T. harzianum MM6. Redundancy analysis (RDA) ascertained that soil C/N ratio and nitrate nitrogen were the main factors affecting the soil microbial diversity. The study provided theoretical basis for soil microbial improvement and biological control of cultivated ginseng.
