    摘要:

    植物根际促生菌是最常用的微生物肥料的菌种来源。施氏假单胞菌(Pseudomonas stutzeri)因良好的促进作物生长、提高作物抗逆性及降解有机污染物等特性备受关注。以具有减少农田土壤氧化亚氮排放和良好促生特性的施氏假单胞菌NRCB010为研究对象,明确其菌落形态及生理生化特征;以生物量(以OD600值表示)为评价指标,通过单因素优选法确定发酵基础培养基和碳氮源及其浓度,通过正交试验设计和极差分析优化无机盐成分,通过响应面分析法优化发酵培养基配方。结果表明,NRCB010菌落呈圆形、皱褶状、黄色,革兰氏染色阴性,菌体短杆状、长度约2 μm、无芽孢;利用多种碳源、氨基酸和羧酸,具有还原力。改良的金氏B(King′s B)培养基为NRCB010发酵较好的基础培养基;以15 g/L糖蜜+葡萄糖(1∶1,W/W)为碳源、以25 g/L牛肉浸粉为氮源时NRCB010的生物量最高。最优无机盐组合为KH2PO4 1.5 g/L、 MgSO4·7H2O 1.5 g/L和NaCl 2.0 g/L。优化后的最优发酵培养基为糖蜜7.1 g/L、葡萄糖8.5 g/L、牛肉浸粉22.9 g/L、MgSO4·7H2O 1.3 g/L、NaCl 2.0 g/L和KH2PO4 1.0 g/L。使用此优化培养基发酵36 h,NRCB010发酵液的OD600值为11.09,有效活菌数为2.87×1010 cfu/mL。

    Abstract:

    Plant growth promoting rhizobacteria are the most common source of microbial fertilizers,while pseudomonas stutzeri has attracted much attention because it can promote the growth and stress resistance of crops,and degrade organic pollutants as well. In the study,NRCB010,a pseudomonas stutzeri that can reduce the emission of nitrous oxide from farmland soil and promote the growth of crops,was taken as the object,for which the colonial morphology,physiological and biochemical characteristics were clarified. With the biomass (represented as OD600 value) of NRCB010 as the index for evaluation,the fermentation basal medium,carbon and nitrogen sources,and the corresponding concentrations were determined by univariate optimum seeking method,the composition of inorganic salts was optimized by orthogonal experimental design and range analysis,and the formula for fermentation medium was optimized by a response surface analysis. The results showed that NRCB010 colony was round,wrinkled,yellow,Gram-negative,with short rod-shaped thallus about 2 μm long and without spores. With a reducing power,it utilized a variety of carbon sources,amino acids and carboxylic acids. The modified King’s B medium was a good basal medium for NRCB010 fermentation,and the biomass of NRCB010 was the highest when 15 g/L molasses + glucose (1∶1,W/W) was used as the carbon source and 25 g/L beef extract powder as the nitrogen source. The composition of the optimal inorganic salts was as follows:1.5 g/L KH2PO4,1.5 g/L MgSO4·7H2O and 2 g/L NaCl. After optimization,the optimal fermentation medium was obtained as follows:7.1 g/L molasses,8.5 g/L glucose,22.9 g/L beef extract powder,1.3 g /L MgSO4·7H2O,2.0 g /L NaCl and 1.0 g/L KH2PO4. After 36 h of fermentation by this optimized medium,the OD600 value of NRCB010 fermentation broth was 11.09. The corresponding effective viable count is 2.87×1010 cfu/mL.
