    摘要:

    筛选获得具有氧化亚氮(N2O)还原活性及植物促生特性的细菌,为新型微生物肥料的研发与应用提供微生物资源。通过固氮培养基筛选以及含硝酸钠和琥珀酸钠的营养肉汤培养基纯化,从水塘芦苇根际土中分离得到一株细菌YSQ030;通过形态观察、16S rRNA 基因序列和生理生化特征分析,对菌株进行鉴定;采用无氮反硝化培养基测定菌株的N2O 还原能力;通过纯培养试验测定菌株的促生特征并通过水培试验研究其对水稻的促生效应。结果显示,菌株YSQ030 为反硝化无色杆菌(Achromobacter denitrificans),含nosZ 基因;在30℃、转速为200r/min、氧气浓度为0% 的前提下,还原N2O 的效率为66.3%;分泌吲哚乙酸、具有溶磷和产生1- 氨基环丙烷-1-羧酸(ACC)脱氨酶的能力;温室水培试验结果表明,与未接菌的对照组相比,接种YSQ030 菌液显著提高了水稻幼苗的地上部鲜重和根鲜重,分别增加了48.3% 和37.6%。本研究表明,YSQ030 具有明显的N2O 还原能力,同时对水稻生长具有显著的促进作用,可为进一步构建具有N2O 还原能力的生物肥料提供良好的种质资源。

    Abstract:

    Aimed to screen bacteria with nitrousoxide(N 2O)-reducing and plant growth-promoting abilities that provide microbial resources for the development and application of new biofertilizers. A bacterial strain named YSQ030 was isolated from the rhizosphere soil of common reed grown in apond through azotobacter mannitol agarplate,and purified on nutrient broth media containing sodium nitrate and sodium succinate. The strain was further characterized through morphologicalobservation,16S rRNA genesequencing,and physiological and biochemical characteristics analyses. The N2O-reducing activity of this strain was measured in a nitrogen-free denitrification medium.Meanwhile,the plant growth-promoting characteristics of this strain was determined under pure culture condition.Moreover,the plant growth-promoting effect on rice was determined under hydroponic condition. The results showed that the strain YSQ030 was a Achromobacter denitrificans possessing nosZ gene that encodes N2O reductase. The N2O reduction efficiency was 66.3% when YSQ030 was cultured in vial with 0% of oxygen concentration in headspace at 30℃ and 200 r/min. The strain YSQ030 could produce indole-3-aceticacid,solubilizephosphate,and synthesize 1-aminocyclopropane-1-carboxylate deaminase. The inoculation with YSQ030 significantly increased the fresh weight of rice seedlings and roots by 48.3% and 37.6% compared with the non-inoculatedcontrol,respectively,in a greenhouse hydroponic experiment. This study suggests that the strain YSQ030 has promising N2O-reducing ability and plant growth-prompting effect onrice,thereby providing invaluable microbial resources for the development of biofertilizer with N2O-reducing ability.
