    摘要:

    从新疆克拉玛依油田作业区周围选择石油污染及盐渍化的 3 个区域采集梭梭根际土壤,测定土壤理化性质;分别以原油与 1- 氨基环丙烷 -1- 羧酸(1-amino-cyclopropane-1-carboxylic acid,ACC)作为唯一碳源和氮源,从土样中分离出两株耐盐碱能力强、ACC 脱氨酶活力较高、原油降解能力较强的菌株。菌株 S8-1 和 W8-4 的 ACC 脱氨酶比活力分别为(0.17±0.06)和(0.77±0.08)U/mg,W8-4 的酶比活力显著高于 S8-1。通过乳化性能的测定,得出菌株 W8-4 和 S8-1 的乳化率分别为(54.92±5.04)% 和(49.59±3.69)%,表明其具有增溶作用,可降低油水界面张力。7 d 的降解试验结果显示,两株菌对原油的降解率分别为 52.82% 和 41.38%。经形态学观察、生理生化鉴定和 16S rDNA 序列分析鉴定 S8-1 为格氏假单胞菌(Pseudomonas gessardii)菌株,W8-4 为假单胞菌属(Pseudommonas sp.)菌株。通过研究不同环境条件对菌株原油降解效果的影响发现,当降解时间为 7 d, 摇床转速为 180 r/min 时,菌株 S8-1 的最佳降解条件是接种量为 4%,初始 pH 为 7,培养温度为 30℃,原油浓度为 0.5 g/L;菌株 W8-4 的最佳降解条件是接种量为 5%,初始 pH 为 8,培养温度为 30℃,原油浓度为 2.5 g/L;此时,两菌株对原油的降解率分别为 55.16% 和 57.89%。促生能力的测定结果表明,两株菌均具有生物固氮和溶磷能力。因此,这两株菌可为将来盐渍化石油污染土壤的治理提供有价值的菌种资源。

    Abstract:

    The rhizosphere soil of Haloxylon ammodendron was collected from 3 areas of petroleum pollution and salinization around Karamay oilfield in Xinjiang,and physicochemical properties of soil were determined. Using crude oil and 1-amino-cyclopropane-1-carboxylic acid(ACC)as the only carbon and nitrogen source,respectively,two strains with strong saline-alkali tolerance,high ACC deaminase activity and strong crude oil degrading ability were isolated from soil samples. The specific activity of ACC deaminase in strains S8-1 and W8-4 was(0.17±0.06)and(0.77±0.08) U/mg,respectively. The specific activity of ACC deaminase in strain W8-4 was significantly higher than that in strain S8-1. Through the determination of emulsification property,it was found that the emulsification rates of strains W8-4 and S8-1 were(54.92±5.04)% and(49.59±3.69)%,respectively,indicating that they had solubilization and could reduce the interfacial tension of oil and water. The results of 7-day degradation test showed that the crude oil degradation rates of the two strains were 52.82% and 41.38%, respectively. After morphological observation,physiological and biochemical identification and 16S rDNA sequence analysis, S8-1 was identified as the strain of Pseudomonas gessardii and W8-4 was identified as the strain of Pseudommonas sp.. By studying the influence of different environmental conditions on the degradation effects of crude oil of the strain,it was found that when the degradation time was 7 days and the rotating speed of the shaker was 180 r/min,the optimal degradation conditions of strain S8-1 were 4% inoculation amount,initial pH 7,30℃ culture temperature and 0.5 g/L crude oil concentration. The optimum degradation conditions of strain W8-4 were 5% inoculation amount,initial pH 8,30℃ culture temperature and 2.5 g/L crude oil concentration. At this time,the crude oil degradation rates of the two strains were 55.16% and 57.89%,respectively. The results of growth promoting ability showed that both strains had the ability of nitrogen fixation and phosphorus solubilization. Therefore,these two strains can provide valuable bacterial resources for the treatment of salinized petroleum polluted soil in the future.
