Effects of zinc fertilizer basal application on the zinc-related soil microbial community structure of potato rhizosphere in dryland

    摘要:

    为研究基施锌肥对旱地马铃薯根际与锌相关土壤微生物群落结构的影响,针对不同锌肥施用量,设置基施ZnSO4·7H2O 0 kg·hm-2(对照)、30 kg·hm-2和 60 kg·hm-2共3个浓度处理。各处理采用Illumina Mi-Seq测序研究锌肥在不同施肥水平下对马铃薯根际土壤微生物数量及群落组成的影响。得到了1054207个有效序列,主要隶属于14门、22纲、56目、94科、210属,在门水平上,变形菌门(Proteobacteria)和浮霉菌门(Planctomycetes)为主要优势类群;属水平以贪噬菌属(Variovorax)和柄杆菌属(Caulobacter)为优势属。随着基施锌肥含量(0~60 kg·hm-2)的增加,贪噬菌属的相对丰度显著提高15.45%,假单胞菌属(Pseudomonas)和马赛菌属(Massilia)的相对丰度差异不显著,只提高1.12%和1.08%,黄单胞菌属(Xanthomonas)和溶杆菌属(Lysobacteria)的相对丰度显著降低4.09%和1.19%;同时微生物菌群数和Chao1指数显著下降,不施锌肥马铃薯根际土壤微生物菌群数最多,施锌60 kg·hm-2土壤微生物菌群数最少;土壤微生物群落结构相似度在较低水平上分为两类,施锌肥样本单独聚成一组,不施锌肥样本聚为另一组,施锌土壤的微生物群落结构明显区别于不施锌肥土壤类型,基施锌肥60 kg·hm-2对土壤微生物菌群群落结构有较大的影响。适量施用锌肥对马铃薯根际土壤微生物的种类和数量有促进效应,但高剂量的锌肥对土壤中酶活性有很强的抑制作用,可破坏微生物种群,降低微生物活性,抑制微生物生长,进而影响土壤质量和马铃薯产量。该结果可为种植马铃薯合理施锌对土壤微生物多样性的研究提供一定的科学依据。

    Abstract:

    To study the effect of zinc fertilizer basal application on the zinc-related soil microbial community structure of potato rhizosphere in dryland,three concentrations of ZnSO4·7H2O,namely 0(control),30 and 60 kg·hm-2 were set up for different levels of zinc fertilizer application.Illumina Mi-Seq sequencing was used to study the effects of zinc fertilizer on the number and community composition of microorganisms in potato rhizosphere soil under different fertilization levels.1054207 valid sequences were obtained,which mainly belonged to 14 phyla,22 classes,56 orders,94 families,and 210 genera.At the phylum level,Proteobacteria and Planctomycotes were the main dominant groups;At the genus level,the dominant genera was Variovorax and Caulobacter.As the basal zinc fertilizer content(0-60 kg·hm-2)increased,the relative abundance of Variovorax significantly increased by 15.45% and the relative abundance of Pseudomonas and Massilia did not differ significantly,increasing by only 1.12% and 1.08%,respectively.At the same time,the number of microbial communities and Chao1 index significantly decreased,with the highest number of microbial communities in the rhizosphere soil of potatoes without zinc fertilizer,and the lowest number of microbial communities in the soil with zinc fertilizer of 60 kg·hm-2.The similarity of soil microbial community structure could be divided into two categories at a lower level.Samples treated with zinc fertilizer were clustered into a separate group,while samples treated without zinc fertilizer were clustered into another group.The microbial community structure of soil treated with zinc fertilizer was significantly different from that of soil without zinc fertilizer.Basal application of 60 kg·hm-2 zinc fertilizer had a significant impact on the microbial community structure of soil.Moderate application of zinc fertilizer had a promoting effect on the types and quantities of microorganisms in potato rhizosphere soil,but high doses of zinc fertilizer had a strong inhibitory effect on enzyme activity in the soil,which could destroy microbial populations,reduce microbial activity,inhibit microbial growth,and ultimately affect soil quality and potato yield.This result provided a scientific basis for the study of the effects of reasonable zinc application on soil microbial diversity in potato cultivation.
