城市绿地土壤解磷菌的筛选及促生效果分析

Phosphate solubilization bacteria screening from urban green space soil and their growth promoting effect analysis

    摘要:

    为了从城市绿地土壤环境中筛选解磷菌,明确其菌属,并从中获得具有促生效果的优势解磷菌株。以园林绿地土壤为对象,通过解磷圈观察从筛选平板上获取解磷菌株,分析16S rDNA片段序列同源性并构建系统发育树。利用钼锑抗显色法检测菌株的解磷能力,同时测定其培养基pH。挑选3个解磷能力强的菌株,对应T1、T2和T3处理组,设置未接菌对照(CK),通过矮牵牛栽培试验检验其促生效果。结果表明,从绿地土壤中筛选出的29株解磷菌分属Kluyvera、Pantoea、Enterobacter、Burkholderia、Paraburkholderia和Ochrobactrum属。菌株的解磷能力分布在8.67~632.62 mg/L之间,对应培养基的pH分布在6.38~4.42之间,二者存在明显的负相关(R2=0.8775)。3株优势解磷菌能显著提升矮牵牛的冠幅、叶片数、花芽数和干重,对株高和SPAD无明显作用。另外,T3菌株能显著提升根体积,T2和T3菌株均能提升植株地上和地下部分磷含量,T1菌株则对上述生长指标无明显促进作用。综合来看,绿地土壤中获得的29株解磷菌酸化培养基的能力在一定程度上能反映其解磷能力。挑选的3株优势菌均对矮牵牛有促生效果,解磷能力最强的T3菌株表现最佳,具备进一步开发为菌剂的潜力。

    Abstract:

    The phosphate-solubilizing bacteria from the soil environment of urban green space were screened and their genera were identified,in order to obtain dominant phosphate-solubilizing strains with growth-promoting effects.Phosphate-solubilizing bacteria from screening plates were obtained by observing the phosphate-solubilizing halo,and the phylogenetic tree was constructed by analysing the homology of 16S rDNA fragments.The phosphate solubilization ability of the strains was quantified with molybdenum antimony anti-coloring method,and pH of the medium was also measured.Selecting three strains with strong phosphate solubilization ability,corresponding to T1,T2 and T3 treatments,to test their growth promoting effect through Petunia hybrida cultivation experiment,non-inoculated strain treatment(CK) was set as control.The result showed that 29 phosphate-solubilizing bacteria were screened from the green space soil,and they belonged to Kluyvera,Pantoea,Enterobacter,Burkholderia,Paraburkholderia and Ochrobactrum genera.The phosphate solubilization ability of the strains ranged from 8.67 to 632.62 mg/L,and the corresponding pH distribution of the media ranged from 6.38 to 4.42.There was a significant negative correlation between them(R2= 0.8775).The three strains significantly increased the crown width,leaf number,flower bud number,and dry weight of P. hybrida,but had no significant effect on plant height and SPAD.T3 strain significantly increased root volume,and T2 and T3 strains both increased phosphorus content in the aboveground and underground parts of the plant,while T1 strain had no significant promoting effect on the above growth indicators.Overall,the ability of the 29 phosphate-solubilizing bacteria to acidify the culture media could reflect their phosphate solubilizing ability to a certain extent.The three selected strains all had growth promoting effects,and the T3 strain with the strongest phosphate solubilization ability performed the best and possessed the potential for further development as a microbial fertilizer.
