    摘要:

    从沼渣堆肥中筛选获得两株具有促生功能的枯草芽孢杆菌,研发制得液体菌剂与固体菌剂,通过田间微区试验分析菌剂对大葱植株生长、产量和品质的影响,并利用高通量测序对大葱根际土壤细菌进行物种多样性分析。结果表明,两株枯草芽孢杆菌BNS1和BNS2均具有固氮、解磷、解钾、产吲哚乙酸和产铁载体能力。制得的两种剂型的菌剂均能有效促进大葱生长,其中施用固体菌剂效果更佳,与不施菌剂相比,固体菌剂处理大葱株高、鲜重、假茎高度、假茎直径分别显著增加了14.75%、15.50%、15.74%、24.24%,植株氮素、磷素、钾素积累量分别显著增加了16.18%、35.46%、33.16%,假茎可溶性糖含量、可溶性蛋白含量和维生素C含量分别显著增加了28.11%、29.03%和10.89%。施用两种剂型的菌剂均影响了大葱根际土壤微生物群落结构,增加了节杆菌属(Arthrobacter)、鞘氨醇单胞菌属(Sphingomonas)、芽孢杆菌属(Bacillus)、链霉菌属(Streptomyces)和假单胞菌属(Pseudomonas)等有益微生物的相对丰度;有益微生物的相对丰度与土壤环境因子具有显著的相关性,其中土壤有效磷和速效钾是影响大葱根际微生物群落组成的关键因素,土壤速效钾含量与芽孢杆菌属菌群的相对丰度呈显著正相关,说明菌剂发挥着潜在土壤解钾功能。研究结果有助于全面了解枯草芽孢杆菌菌剂的实际应用价值,为菌剂在大葱等经济作物优质高产栽培中的应用提供理论参考和技术指导。

    Abstract:

    Two strains of Bacillus subtilis with growth-promoting function were screened from biogas slurry compost,and the liquid and solid inoculants were developed and prepared.The effects of these inoculants on the growth,yield and quality of scallion plants were analyzed through a field micro-plot experiment.The species diversity of bacteria in the rhizosphere soil of scallion was analyzed by high-throughput sequencing.The results showed that the two strains of Bacillus subtilis BNS1 and BNS2 had the ability of nitrogen fixation,phosphate solubilization,potassium solubilization,IAA production and siderophore production.The two kinds of inoculant effectively promoted the growth of scallion,and the effect of solid inoculant was better.Compared with CK,the plant height,fresh weight,false stem height and false stem diameter of scallion treated with solid inoculant significantly increased by 14.75%,15.50%,15.74% and 24.24%,respectively.The accumulation of nitrogen,phosphorus and potassium increased by 16.18%,35.46% and 33.16%,respectively.The contents of soluble sugar,soluble protein and vitamin C in pseudostem increased by 28.11%,29.03% and 10.89%,respectively.The microbial community structure in rhizosphere soil of scallion was affected by the application of two types of inoculants.The relative abundance of beneficial microorganisms such as Arthrobacter,Sphingomonas,Bacillus,Streptomyces and Pseudomonas was increased.The relative abundance of beneficial microorganisms was significantly correlated with soil environmental factors,in which soil available phosphorus and available potassium were the key factors affecting the composition of rhizosphere microbial community of scallion,and soil available potassium content was significantly positively correlated with the relative abundance of Bacillus flora,indicating that inoculant played a potential role in soil potassium solubilization.The results were helpful to understand the practical application value of Bacillus subtilis,and that would provide theoretical reference and technical guidance for the application of Bacillus subtilis in the high quality and high yield cultivation of cash crops such as scallion.
