铅胁迫下北苍术幼苗生长生理响应及耐铅性分析

    摘要:

    探究不同土壤浓度铅(Pb)胁迫下北苍术幼苗生长、生理和Pb含量的响应特征,以期为北苍术安全生产提供理论参考。试验在不同时间(0、5、10、15、25 d)挖取施入不同土壤浓度Pb(0、66.67、133.33、200.00和266.67 mg·kg-1)处理的北苍术幼苗,分析北苍术幼苗在生长、生理和Pb含量等方面的响应特征。结果表明:不同浓度Pb处理条件下,北苍术幼苗生长相关指标(叶片数、叶长、叶宽、须根长、须根直径、须根数、地上和地下部分干重)、耐性系数和生理相关指标(可溶性糖含量、总蛋白含量、叶绿素含量、根系活力、过氧化物酶活性和过氧化氢酶活性)随Pb浓度增大均呈先升高后下降的趋势。脯氨酸含量、丙二醛含量和超氧化物歧化酶活性随Pb浓度增大呈逐渐升高的趋势。在Pb浓度66.67 mg·kg-1处理5~10 d、133.33 mg·kg-1处理5 d时,北苍术幼苗地下部分Pb含量<5 mg·kg-1,符合《中华人民共和国药典》质量安全标准;地下部分富集系数范围为0.04~0.66;转运系数范围为1.49~8.53;相关性分析表明,生长指标与部分生理指标间呈显著或极显著正相关,与部分生理指标和Pb含量之间呈显著或极显著负相关。主成分分析表明过氧化物酶活性、根系活力、须根长、叶绿素含量、丙二醛含量、地上和地下部分Pb含量可以作为北苍术幼苗响应Pb胁迫的关键指标。

    Abstract:

    This study aimed to investigate the response characteristics of growth,physiology and lead(Pb)content in the seedlings of Atractylodes chinensis under different soil Pb concentrations,providing a theoretical reference for the safe production of Atractylodes chinensis. Seedlings of Atractylodes chinensis were treated with various soil Pb concentrations(0,66.67,133.33,200.00 and 266.67 mg·kg-1)at different time points(0,5,10,15,25 days)were excavated,and their growth,physiological responses and Pb content were analyzed. The results indicated that under different Pb concentrations,the growth-related indices(leaf number,leaf length,leaf width,fibrous root length,fibrous root diameter,fibrous root number,dry weight of above- and below-ground parts),tolerance coefficients and physiological indices(soluble sugar content,total protein content,chlorophyll content,root activity,peroxidase activity and catalase activity)of Atractylodes chinensis seedlings initially increased and then decreased with the increase in Pb concentration. Meanwhile,proline content,malondialdehyde content and superoxide dismutase activity were gradually increased with the increase in Pb concentration. When treated with 66.67 mg·kg-1 Pb for 5-10 days or 133.33 mg·kg-1 Pb for 5 days,the Pb content in the below-ground parts of Atractylodes chinensis seedlings was less than 5 mg·kg-1,meeting the quality and safety standards of Pharmacopoeia of the People’s Republic of China. The bioconcentration factor ranged from 0.04 to 0.66,and the translocation factor ranged from 1.49 to 8.53 in the below-ground parts. Correlation analysis revealed significant or extremely significant positive correlations between growth indices and some physiological indices,while significant or extremely significant negative correlations were observed between growth indices and other physiological indices as well as Pb content. Principal component analysis indicated that peroxidase activity,root activity,fibrous root length,chlorophyll content,malondialdehyde content and Pb content in both above- and below-ground parts could serve as key indicators for the response of Atractylodes chinensis seedlings to Pb stress.
