    摘要:

    为探明3种微生物肥料对穿心莲根际土壤微生态结构的影响,以穿心莲为试验材料,设置高浓度光合细菌液肥(GH)、寡糖液肥(GT)、小球藻液肥(XQ)和清水(CK)的4个处理,采用高通量测序技术分析穿心莲根际微生物对3种肥料处理的生态响应及其引起的群落结构变化情况。结果显示:各施肥处理组中全氮含量均出现显著提升(P<0.05),其中施用小球藻肥后全氮含量提升了52.17%,作用效果XQ>GH>GT。碱解氮含量也出现微量提升,作用效果GT>XQ>GH。施用光合细菌肥料和寡糖肥料均提高了土壤中速效钾含量,且前者达到显著水平(P<0.05)。施用菌肥后土壤中有效磷含量均出现一定程度下降,表现为XQ>GT>GH,但均未达到显著水平。冗余分析结果显示:穿心莲根际土壤养分因子中,速效养分(速效钾、有效磷)在较大程度上解释了土壤微生物群落结构的变化情况。穿心莲4个处理组根际土样中检测出主要优势细菌门为变形菌门(Proteobacteria)、酸杆菌门(Acidobacteriota)、拟杆菌门(Bacteroidota)等,主要优势真菌门为子囊菌门(Ascomycota)、担子菌门(Basidiomycota)。主坐标分析表明根际微生物群落结构因微生物肥料的使用产生了显著性变化(P<0.05)。研究表明,施用微生物肥料能显著影响根际菌群结构,在一定程度上降低了土壤中真菌与细菌OTU数量的比值,增加土壤肥力并促进植物对必需营养元素的吸收。研究结果为穿心莲种植遴选适宜微生物肥料种类以及益生菌群构建提供了理论参考。

    Abstract:

    To explore the effects of three kinds of biofertilizers on the microenvironmental structure of the rhizosphere soil of Andrographis paniculata,an experiment was conducted with the Andrographis paniculata rhizosphere soil treating with high concentration photosynthesizing bacterial liquid fertilizer(GH),oligosaccharides liquid fertilizer(GT),Chlorella liquid fertilizer(XQ),and clear water(CK),respectively. The Illumina high-throughput sequencing technology was used to analyze the ecological responses of the microorganisms and the community structure of Andrographis paniculata’s rhizosphere soil to the three different kinds of fertilizer treatments. The results showed that the total nitrogen content in each fertilization group were significantly increased(P<0.05)with the effect of XQ>GH>GT,especially the total nitrogen content was increased by 52.17% after the application of Chlorella fertilizer. The alkaline hydrolysis nitrogen content also showed a slight increase,with the effect of GT>XQ>GH. The application of photosynthetic bacteria fertilizer and oligosaccharide fertilizer both increased the content of available potassium in the soil,and the former reached a significant level(P<0.05). After the application of microbial fertilizer,the content of available phosphorus in the soil decreased to a certain extent,showing XQ>GT>GH,but did not reach a significant level. The results of redundancy analysis showed that among the inter-root soil nutrient factors of Andrographis paniculata,the available potassium and available phosphorus explained to a greater extent the variations in soil microbial community. The main dominant bacterial phyla detected in the rhizosphere soil samples of Andrographis paniculata in the treatments were Proteobacteria,Acidobacteriota and Bacteroidota,etc. The main dominant fungal phyla were Ascomycota and Basidiomycota. The principal coordinate analysis showed that the structure of rhizosphere soil microbial community was significantly changed(P<0.05)due to the application of biofertilizer. The study showed that the application of biofertilizer could significantly affect the structure of the rhizosphere microbiota,reduce the ratio of fungal and bacterial OUT counts in the soil,increase soil fertility and promote the uptake of essential nutrients by plants. This study could provide a theoretical reference for the selection of suitable bacterial fertilizer and the construction of probiotic communities in Andrographis paniculata cultivation.
