北苍术根际土壤微生物与挥发性物质含量相关性研究

Correlation between rhizosphere soil microorganisms and volatile substances contents of Atractylodes chinensis

    摘要:

    苍术历来是防治疫病之要药,但目前野生资源破坏严重、人工栽培质量良莠不齐。基于细菌16S rDNA的V3+V4区和真菌ITS区,应用Illumina NovaSeq高通量测序技术并结合生物信息学比较野生与人工栽培北苍术根际土壤细菌和真菌群落结构,并与北苍术根茎内挥发性物质进行关联分析,旨在揭示土壤微生态环境对北苍术挥发性物质含量的影响。结果表明,两处理土壤的优势细菌门为变形菌门、放线菌门和酸杆菌门,在野生土壤中相对丰度较大的细菌有变形菌门、酸杆菌门、α-变形菌纲、NC10纲、根瘤菌目、鞘氨醇单胞菌科等;两处理土壤的优势真菌门为子囊菌门、担子菌门和被孢霉门,在野生土壤中相对丰度较大的真菌有担子菌门、伞菌纲、古根菌目、柔膜菌科、Solicoccozyma aeria等;野生土壤中参与土壤碳循环的化能异养细菌和有益于植物生长的青霉菌属真菌丰度较高,在栽培土壤中病原真菌镰孢菌属和木贼镰孢菌则丰度较高。相关性分析结果表明,在野生土壤中相对丰度较大的大多数微生物与不同类型的挥发性物质含量均有显著相关性。本研究对野生和人工栽培北苍术根际土壤微生物群落结构的明确,为实现野生北苍术根际有益微生物的功能挖掘和生物肥料研究、改善栽培北苍术土壤微环境、提高药效成分含量提供理论依据。

    Abstract:

    Atractylodes chinensis has always been an important medicine for disease prevention and control,but the wild resources are seriously damaged and the quality of artificial cultivation is uneven. Based on the V3+V4 region of 16S rDNA of bacteria and ITS region of fungi,Illumina NovaSeq high-throughput sequencing technology combined with bioinformatics were applied to compare the community structure of bacteria and fungi in rhizosphere soil of wild and cultivated Atractylodes chinensis. The aim of correlation analysis between rhizosphere soil microorganisms and volatile substances content was to reveal the influence of soil microecological environment on volatile substances in Atractylodes chinensis. The results showed that the dominant bacterial phyla of the two treatments were Proteobacteria,Actinomycetes and Acidobacteria. The relatively abundant bacteria in wild soil were Proteobacteria,Acidobacteria,α-Proteobacteria,NC10,Rhizobiales,Sphingomonadaceae,etc. The dominant fungal phyla in the two treatments were Ascomycota,Basidiomycota and Mortierellomycota. Fungi with relatively high abundance in wild soil included Basidiomycota,Agaricomycetes,Archaeorhizomycetales,Helotiaceae,Solicoccozyma aeria,etc. Chemoheterotrophic bacteria involved in soil carbon cycle and fungi of Penicillium were abundant in wild soil,while in cultivated soil,pathogenic fungi of Fusarium and Fusarium equiseti were abundant. The results of correlation analysis showed that most of the relatively abundant microorganisms in wild soil were significantly correlated with the contents of different types of volatile substances. This study clarified the microbial community structure of rhizosphere soil of wild and artificially cultivated Atractylodes chinensis,providing theoretical basis for functional exploration of beneficial microorganisms in rhizosphere of wild Atractylodes chinensis and biofertilide research,improving the soil microenvironment and increasing the content of medicinal ingredients of cultivated Atractylodes chinensis.
