Influence of different fertilization treatments on the characteristics and diversity of soil bacterial community in strawberry orchards in dry-hot valley

    摘要:

    为探明有机肥对草莓园土壤细菌群落的影响,为干热河谷区优化草莓园施肥提供理论依据。以蚯蚓肥 (TA)、油枯肥(TB)、蚯蚓肥 + 油枯肥配施(TC)以及不施肥作为对照(CK)为试验处理,采用 Illumina Miseq 高通量测序技术,分析施用有机肥对土壤细菌群落丰度、多样性及其群落组成特征。结果表明,不同施肥处理均能提高土壤理化性质和土壤酶活性,其中,TA 处理显著提高了土壤 pH 值,TB 处理对碱解氮、有效磷和速效钾含量的提升作用最明显,TB 处理下土壤微生物量碳、微生物量氮、脲酶活性、酸性磷酸酶活性、过氧化氢酶活性均显著升高。不同施肥处理均显著影响了土壤细菌群落多样性和群落结构,其中,TA 处理显著提高了土壤 Chao 指数、Shannon 指数,优化了土壤细菌群落结构,使细菌共生群落变得丰富且更稳定;TB 处理则显著降低了土壤 Chao 指数、Shannon 指数,且对细菌群落结构的影响较小。冗余分析表明,放线菌门(Actinobacteriota)、绿弯菌门(Chloroflexi)与 pH、全钾呈正相关,变形菌门(Proteobacteria)、拟杆菌门(Bacteroidota)与碱解氮、有机质、全氮呈正相关,Firmicutes 与全磷、容重、速效钾、有效磷呈正相关。细菌群落与 pH、有机质、全氮、碱解氮呈极显著正相关(P<0.01)。研究表明,增施蚯蚓肥可以调节 pH 值,提高细菌群落多样性和类群丰度,油枯可以提高土壤肥力,两者配施能够有效改善土壤理化性质,提升土壤养分供应水平,优化细菌群落结构,可优先作为干热河谷区草莓园土壤培肥改良的肥料。

    Abstract:

    The effects of organic fertilizer on soil bacterial community of strawberry garden were explored to provide theoretical basis for optimizing the fertilization of strawberry garden in dry and hot river valley area. Using earthworm fertilizer (TA),oil dry fertilizer(TB),earthworm fertilizer combined with oil dry fertilizer(TC)and no fertilizer as control (CK)as experimental treatments,Illumina Miseq high-throughput sequencing technology was used to analyze the effects of organic fertilizer on soil bacterial community abundance,diversity and community composition. The results showed that different fertilization treatments improved soil physicochemical properties and soil enzyme activities,among which TA treatment significantly increased soil pH value,TB treatment had the most obvious effect on the content of alkali-hydrolyzed nitrogen(AN),available phosphorus(AP)and available potassium(AK). Soil microbial biomass carbon,microbial biomass nitrogen,urease activity,acid phosphatase activity and catalase activity were significantly increased under TB treatment. Different fertilization treatments significantly affected soil bacterial community diversity and community structure. Among them,TA treatment significantly increased soil Chao1 index and Shannon index,optimized soil bacterial community structure,and made the bacterial symbiotic community more abundant and stable. TB treatment significantly reduced soil Chao1 index and Shannon index,and had little effect on bacterial community structure. Redundancy analysis(RDA) showed that Actinobacteriota and Chloroflexi were positively correlated with pH and total potassium,while Proteobacteria and Bacteroidota were positively correlated with AN,organic matter(OM)and total nitrogen(TN). Firmicutes were positively correlated with total phosphorus,bulk density,AK and AP. The bacterial community was positively correlated with pH, OM,TN and AN(P<0.01). The results showed that increased application of earthworm fertilizer could adjust pH value, improve the diversity and abundance of bacterial community,oil dry fertilizer could improve soil fertility,and its mixed application could effectively improve soil physical and chemical properties,enhance soil nutrient supply level,and optimize the structure of bacterial community. It could be used as a preferred fertilizer for soil fertilizer improvement in strawberry garden in dry and hot river valley area.
