    摘要:

    探究钾长石土壤调理剂与不同用量的胶质芽孢杆菌配施对翠碧一号烤烟根系生长及烤烟根际土壤细菌多样性和群落结构的影响,为福建省龙岩市长汀烟区土壤改良及提高土壤微生态环境质量提供参考依据。采用大田试验和高通量测序技术,分析施加不同用量的胶质芽孢杆菌菌剂(T1、T2、T3 处理)、钾长石土壤调理剂(T4 处理)、钾长石土壤调理剂 + 不同用量的胶质芽孢杆菌菌剂(T5、T6、T7 处理)的烤烟根系发育、根际土壤理化性质、酶活性及微生物群落结构和功能。相较于常规施肥(CK),随着生育期的推进,钾长石土壤调理剂与菌剂单施或混施均有提高烤烟根系活力、改善根系形态的作用,且以 T6 和 T7 处理作用效果显著;各处理土壤 pH 逐渐上升,有机质、碱解氮及有效磷含量均呈先升后降的趋势,且在旺长期达到最大,速效钾含量逐渐下降至打顶期后缓慢上升,不同生育期均以 T6 和 T7 处理的速效钾含量较高;不同处理硝酸还原酶、脲酶、蔗糖酶和过氧化氢酶含量均在旺长期达到最大,以 T6 处理酶活性最高;施用菌剂可提高根际土壤微生物群落丰度及多样性指数,T2 和 T6 处理以铁氧化属(Sideroxydans)、硝化刺菌属 (MBNT15)和 BSV26 为优势菌属,丰度分别为 3.70%、3.51%、3.25% 和 3.89%、3.26%、3.21%;在细菌群落 Alpha 多样性分析中,各处理的 Shannon 指数、Simpson 指数、Chao1 指数、Ace 指数和 PD 指数排序均表现为 T6>T2>T4>CK,改变了根际土壤的物种丰度。钾长石土壤调理剂与菌剂单施或混施均能够促进烤烟根系的发育,提高根际土壤养分及酶活性,增加土壤细菌多样性及改良微生物群落结构,其中,以 T6 处理作用效果最佳。

    Abstract:

    The effects of potassium feldspar soil conditioner combined with different dosage of Bacillus mucilaginosus on the root growth of CB-1 flue-cured tobacco and the bacterial diversity and community structure in the rhizosphere soil of fluecured tobacco were explored,so as to provide reference for soil improvement and soil microecological environment quality improvement in Changting tobacco-growing area. Using field experiment and high-throughput sequencing technology,the root development,rhizosphere soil physical and chemical properties,enzyme activities,microbial community structure and function of flue-cured tobacco with different dosage of Bacillus mucilaginosus(T1,T2 and T3 treatments),potassium feldspar soil conditioner(T4 treatment)and potassium feldspar soil conditioner combined with different dosage of Bacillus mucilaginosus(T5,T6 and T7 treatments)were analyzed. Compared with conventional fertilization(CK),single or mixed application of potassium feldspar soil conditioner and microbial inoculum could effectively improve root activity and root morphology of flue-cured tobacco,and the effects of T6 and T7 treatments were significant. The pH of soil in each treatment was gradually increased,the contents of organic matter,alkali-hydrolyzable nitrogen and available phosphorus were all increased first and then decreased,and reached the maximum in the prosperous period. The content of available potassium gradually was decreased to the topping stage and slowly increased,and the content of available potassium in T6 and T7 treatments was higher in different growth stages. The contents of nitrate reductase,urease,sucrase and catalase in different treatments all reached the maximum in the flourishing period,and the enzyme activity in T6 treatment was the highest. The application of microbial agents could improve the abundance and diversity index of rhizosphere soil microbial community. In T2 and T6 treatments,SideroxydansMBNT15 and BSV26 were the dominant bacteria,with the abundance of 3.70%,3.51%,3.25% and 3.89%,3.26% and 3.21% respectively. In the analysis of Alpha diversity of bacterial community,the order of Shannon index,Simpson index,Chao1 index,Ace index and PD index of each treatment was T6 > T2 > T4 > CK. Potassium feldspar soil conditioner and microbial inoculum can promote the root development of flue-cured tobacco,improve that nutrient and enzyme activity of rhizosphere soil, increase soil bacterial diversity and improve microbial community structure. Among them,T6 treatment showed the best effect.
