四株高产吲哚乙酸菌株的筛选鉴定及促生应用研究

Screening,identification and growth-promoting application research of four high IAA-producing strains

    摘要:

    现代农业中化肥长期不合理施用直接导致了土壤质量和农产品品质下降的问题,微生物肥料替代是解决这些难题的重要途径。试验从前期实验室保存的芽孢杆菌库中筛选出 9 株高产吲哚乙酸(IAA)的菌株,IAA 分泌量均在 83.27 mg/L 以上,其中 T26 分泌能力最强,为 189.14 mg/L。综合产 IAA 能力、产铁载体能力、抑菌特性等生物学特性,确定 4 株菌(T26、PA8、DY15 和 Ya6)开展进一步研究。菌株鉴定结果表明,T26、PA8、 DY15 初步归属于 Bacillus subtilis,Ya6 初步归属于 Bacillus velezensis。4 株菌均具有产铁载体能力,Ya6 具有较强的镰刀菌属真菌(Fusarium)拮抗能力。促生能力研究结果表明,添加高产 IAA 菌株的育苗基质显著促进了番茄、辣椒和糯玉米的苗期生长。番茄育苗试验中 T26、PA8 和 DY15 促生效果佳,番茄株高、茎粗、地上部鲜重、 地下部鲜重分别较对照(添加等量蒸馏水)提高了 24.75% ~ 51.34%、36.08% ~ 43.67%、21.67% ~ 91.67%、 11.63% ~ 27.91%。4 株菌在低肥力土壤中应用促生效果优于在育苗基质中的应用,添加菌株处理的小白菜株高、 茎粗、叶片数、叶面积、根长、地上部鲜重和地下部鲜重与对照处理呈显著性差异,分别为对照的 1.80 ~ 2.08、 1.38 ~ 1.70、1.50、2.80 ~ 3.69、1.12 ~ 1.72、4.18 ~ 5.85 和 4.45 ~ 6.41 倍。4 株菌均具有良好的促生应用潜力, 将为开发微生物肥料提供优良的菌种资源。

    Abstract:

    The long-term unreasonable application of chemical fertilizers in modern agriculture had directly led to the decline of soil and agricultural product quality. Microbial fertilizer substitution was an important way to solve these problems. Nine high-yielding strains of indole-3-acetic acid(IAA)were screened from the Bacillus library stored in the previous laboratory. The IAA secretion amounts of all strains are above 83.27 mg/L. Among them,T26 has the strongest secretion ability,which is 189.14 mg/L. Based on the comprehensive biological characteristics such as IAA production ability,iron carrier production ability,and antibacterial properties,four strains of bacteria(T26,PA8,DY15 and Ya6)were identified for further research. The results of molecular biological identification showed that T26,PA8,and DY15 belong to Bacillus subtilis, while Ya6 belongs to Bacillus velezensis. All four strains have the ability to produce iron carrier. Ya6 has a strong antagonistic ability against Fusarium. The research results of growth-promoting application ability showed that the seedling substrate added with high-yield IAA strains significantly promoted the seedling growth of tomatoes,peppers and glutinous corn. In tomato seedling experiment,T26,PA8 and DY15 had better growth promotion effects,and the plant height,stem diameter, fresh weight of aboveground and underground of tomato increased by 24.75%-51.34%,36.08%-43.67%,21.67%- 91.67%,11.63%-27.91%,respectively,compared with the control. The application of four strains of bacteria in lowfertility soil demonstrated superior growth-promoting effects compared to their use in seedling substrates. The plant height, stem diameter,number of leaves,leaf area,root length,above-ground fresh weight,and below-ground fresh weight of the treated pak choi plants showed significant differences compared to the control group. Specifically,these parameters were 1.80-2.08,1.38-1.70,1.50,2.80-3.69,1.12-1.72,4.18-5.85,and 4.45-6.41 times greater than those of the control, respectively. All four strains had good growth-promoting application potential,which would provide excellent strain resources for microbial fertilizer.
