The effects of different multiple cropping methods on soil microbial community structure and enzyme activity

    摘要:

    为了分析复种饲草对土壤微生物群落组成及酶活性的影响,采用16S rDNA高通量测序对复种饲草土壤中细菌和真菌群落结构,以及土壤酶活性进行分析。结果表明:(1)复种饲草土壤中微生物多样性不同,复种豌豆的过程中,土壤内生菌存在协同共进,其微生物多样性会增加,Alpha多样性指数表明,土壤中微生物群落的Observed species指数、Simpson指数、Shannon指数和Chao1指数高于复种燕麦和休闲地的土壤中微生物群落的相应指数;(2)复种饲草土壤中的优势细菌和真菌存在差异,运用热图对复种不同饲草的土壤样本属水平微生物群落进行分析,结果表明,复种豌豆后土壤中的细菌Acidibacter、Subgroup_6、Gitt-GS-136等丰度显著提高,土壤中的真菌Fusarium、Cylindrocarpon、Microdochium、Ophiosphaerella、Rhizopus、Lophiostoma、Actinomucor等丰度同样显著提高;(3)复种饲草影响与土壤营养循环相关的酶活性,经过1个生长季节后,随着时间的推进,复种饲草土壤中酸性磷酸酶、蔗糖酶、脲酶活性呈增加的趋势;多酚氧化酶活性呈降低的趋势,以藏青2000、藏青17、喜拉22和13-5171-7作为前茬作物复种豌豆,土壤中土壤脲酶活性分别提高13%、4%、9%和14%,同时土壤中过氧化氢酶活性分别提升4%、3%、6%和6%,综上所述,复种饲草对土微生物的多样性及酶活性具有一定的影响。

    Abstract:

    In order to analyse the effect of multiple cropping forage on soil microbial community composition and enzyme activity,this study used 16S rDNA high-throughput sequencing to analyse the structure of bacterial and fungal communities in multiple cropping forage soil,as well as soil enzyme activity. The results showed that:(1)The microbial diversity in the soil of multiple cropping forage was different,and the microbial diversity increased during the replanting of pea with the presence of soil endophytes in common synergism,and the Alpha diversity index showed that the Observed species index,Simpson index,Shannon index and Chao1 index of the microbial community in the soil were higher than those of the soil of multiple cropping oat and recreational land. The indexes of microbial community in the soil were higher than those in the multiple cropping of oats and recreational land. (2)There were differences in the dominant bacteria and fungi in the soil of multiple cropping of forage grass. Heatmap analysis was used to analyze the genus level microbial communities in soil samples of different forage grasses. The results showed that after multiple cropping of peas,the abundance of bacteria such as Acidibacter,Subgroup-6,Gitt-GS-136 in the soil significantly increased,and the abundance of fungi such as Fusarium,Cylindrocarpon,Microdochium,Ophiosphaerella,Rhizopus,Lophiostoma,Actinomucor in the soil also significantly increased;(3)Multiple cropping of forage could affect enzyme activity related to soil nutrient cycling. After one growing season,with the passage of time,the activities of acid phosphatase,sucrase,and urease in the soil of multiple cropping forage showed an increasing trend;polyphenol oxidase showed a decreasing trend. When peas were replanted with Zangqing 2000,Zangqing 17,Xila 22,and 13-5171-7 as previous crops,soil urease activity increased by 13%,4%,9%,and 14%,respectively. At the same time,soil catalase activity increased by 4%,3%,6%,and 6%,respectively. In summary,intercropping forage had a certain impact on soil microbial diversity and enzyme activities.
