Screening and identification of inter-root potassium-solubilising bacteria of Fritillaria taipaiensis P.Y.Li from different origins and their growth-promoting characteristics

    摘要:

    研究不同产地太白贝母的根际土壤解钾菌的功能菌株和高效菌株,并探究优良菌株的多样性以及对太白贝母的促生作用,为太白贝母植株产量及品质提升奠定基础。采用黄色解钾圈法对解钾菌株进行初筛,采用以钾长石为钾源的硅酸盐液体培养进行菌株解钾能力的定量测定,使用火焰分光光度法测定解钾能力,解钾能力指标筛选出优势菌45株;采用细菌促生能力指标溶磷、产铁、产吲哚乙酸(IAA)能力对菌株进行复筛。通过菌株形态学鉴定、革兰氏染色观察、生理生化实验与16S rDNA测序分析对3株高效促生解钾菌进行鉴定。结果表明:从太白贝母根际土壤样品中共分离得到103株解钾菌,其中45株解钾能力较为优势,通过初筛和复筛得到3株高效解钾菌株K9-4、K2-4、K4-1,分别为蜡状芽孢杆菌(Bacillus cereus)、伯克霍尔德菌(Burkholderia hospita)和枯草芽孢杆菌(Bacillus subtilis),分属于2株芽孢杆菌属和1株伯克霍尔德菌属。解钾和溶无机磷能力最强的菌株为蜡状芽孢杆菌,增钾率达60.32%,对钾长石粉的最高转化量达1.26 mg·L-1、对磷酸三钙转化量达8.85 mg·L-1;产IAA和溶有机磷能力最强的菌株为枯草芽孢杆菌,对卵磷脂的最高转化量达2.78 mg·L-1、产铁指数高达2.42;而伯克霍尔德菌株各项能力均介于其余两株之间。综上,成功筛选出3株高效解钾菌,具有强大的解钾、促生能力。这些菌株为微生物解钾菌肥的开发提供菌种资源,同时也为太白贝母植株的促生肥料选择提供更多选项。

    Abstract:

    The aimed to study the functional and efficient strains of potassium-solubilising bacteria in the rhizosphere soil of Fritillaria taipaiensis from different origins,and to explore the diversity of good strains and their growth-promoting effects on Fritillaria taipaiensis,so as to lay the foundation for the improvement of yield and quality of Fritillaria taipaiensis plants.The yellow potassium-solubilising circle method was used for the initial screening of potassium-solubilising strains,silicate liquid culture with potassium feldspar as potassium source was used for the quantitative determination of potassium-solubilising capacity of the strains,and the potassium-solubilising capacity was determined using the flame spectrophotometer method,and the indicators of potassium-solubilising capacity were screened to find out 45 dominant bacterial strains;and the indicators of the bacterial biochemical capacity,such as the capacity of phosphorus-solubilising,iron-producing and 3-indoleacetic acid(IAA)-producing,were used to conduct the re-screening of bacterial strains.Three strains of highly efficient potassium-solubilising bacteria were identified by strain morphology,Gram staining observation,physiological and biochemical experiments and 16S rDNA sequencing analysis.Results showed that a total of 103 strains of potassium-solubilising bacteria were isolated from the rhizosphere soil samples of Fritillaria taipaiensis,among which 45 strains were more advantageous in potassium-solubilising ability,and three highly efficient potassium-solubilising bacterial strains,K9-4,K2-4,and K4-1 were obtained by primary and re-screening,which were Bacillus cereus,Burkholderia hospita and Bacillus subtilis,respectively,belonging to two strains of Bacillus and one strain of Burkholderia.The strain with the strongest ability to solubilise potassium and inorganic phosphorus was Bacillus cereus,with a potassium enhancement rate of 60.32%,and the highest conversion of potassium feldspar powder up to 1.26 mg·L-1,and the conversion of tricalcium phosphate up to 8.85 mg·L-1;the strain with the strongest ability to produce IAA and solubilise organic phosphorus was Bacillus subtilis,with a highest conversion of lecithin up to 2.78 mg·L-1,and the iron production index was as high as 2.42;while the above functions of Burkholderia cepacia strain were between the remaining two strains.In this study, three strains of high-efficiency potassium-solubilising bacteria with strong potassium-solubilising and life-promoting abilities were successfully screened.These strains could provide strain resources for the development of microbial potassium-solubilising bacterial fertilizers, as well as more options for the selection of pro-biotic fertilizers for Fritillaria taipaiensis plants.
