Comprehensive evaluation of Cd tolerance in oilseed rape with different leaf colors in nearly isogenic lines

    摘要:

    为掌握镉(Cd)胁迫条件下,紫叶油菜与绿叶油菜的耐Cd能力差异,以19-Z10-P紫叶油菜(PR)和19-Z10-G绿叶油菜(GR)为供试材料,开展Cd胁迫的水培试验,设置0 mg·L-1(对照)、30 mg·L-1(处理)两个Cd浓度,分析Cd胁迫后油菜株高、根长、茎叶鲜重、根鲜重、茎叶干重、根干重、叶绿素、花青素、相对含水量、相对电导率、丙二醛、脯氨酸、过氧化氢、可溶性糖、超氧化物歧化酶、过氧化物酶、过氧化氢酶、茎叶Cd含量、根Cd含量、单株提取量、耐Cd指数、转运系数等指标,研究Cd胁迫下两种油菜耐Cd能力的差异。研究发现:(1)与对照相比,Cd处理后,PR、GR的株高、根长、茎叶鲜重、根鲜重、茎叶干重、根干重、叶绿素、花青素、相对含水量及过氧化氢酶活性均有所下降,相对电导率、丙二醛、脯氨酸、过氧化氢、可溶性糖含量及超氧化物歧化酶、过氧化物酶活性则有所上升;上述指标中,GR、PR的株高、茎叶鲜重、叶绿素a、相对含水量、相对电导率变幅相当,叶绿素b、叶绿素a+b和花青素降幅GRPR;(2)PR、GR吸收、转运Cd的能力差别不大,茎叶Cd含量、根Cd含量、单株提取量、Cd转运系数(TF)差异均不显著,Cd耐性指数PR(-0.44)>GR(-0.58);(3)隶属函数法综合评价得分,PR(0.50)>GR(0.41)。Cd胁迫后,PR、GR的生长受抑,光合色素及花青素含量下降,叶片失水皱缩,活性氧增加,膜受到损伤,渗透调节物质增加,超氧化物歧化酶、过氧化物酶活性增加,过氧化氢酶活性下降,PR的Cd耐性高于GR。

    Abstract:

    In order to investigate the differences in Cd tolerance between purple leaf rape(PR)and green leaf rape(GR)under Cd stress conditions,a hydroponic experiment was conducted using 19-Z10-P PR and 19-Z10-G GR as test materials. Two Cd concentrations,0 mg·L-1(control)and 30 mg·L-1(treatment),were set up to analyze the plant height(Ph),root length(Rl),shoot fresh weight(Sfw),root fresh weight(Rfw),shoot dry weight(Sdw),root dry weight(Rdw),chlorophyll(Chl),anthocyanin(Ant),relative water content(Rwc),relative electrical conductivity(Rec),malondialdehyde(MDA),proline(Pro),hydrogen peroxide(H2O2),soluble sugar(Ss),superoxide dismutase(SOD),peroxidase(POD),catalase(CAT),Cd content,single plant extraction amount(Ea),cadmium tolerance coefficient(TI),and transport coefficient(TF)of rapes after Cd stress,and the differences in Cd tolerance between two rape varieties were studied. The results showed that:(1)Compared with CK,after treated with Cd stress,the pH,RL,Sfw,Rfw,Sdw,Rdw,Rwc,Chl,Ant content and CAT activity of PR and GR were decreased,while the Rec,MDA,Pro,H2O2,Ss content and SOD,POD activity were increased. Among the above indicators,the Ph,Sfw,Chla,Rwc,and Rec of GR and PR had similar variations,while the Chlb,Chla+b and Ant variations showed as GRPR;(2)The difference in the ability of PR and GR to absorb and transport Cd was not significant,and the differences of the shoot Cd content,root Cd content,single plant extraction amount,and the translocation factor of Cd were not significant,TI showed as PR(-0.44)>GR(-0.58);(3)The comprehensive evaluation score of the membership function method showed as PR(0.50)>GR(0.41). So,after Cd stress,the growth of PR and GR was inhibited,the content of photosynthetic pigments and anthocyanins decreased,leaves lost water and turned yellow,reactive oxygen species increased,membrane damage occurred,and osmotic regulatory substances increased. The Cd tolerance index and comprehensive Cd tolerance ability of PR were higher than those of GR.
