枝条覆盖下梨园土壤微生态垂直变化分析

    摘要:

    为明确枝条覆盖对梨园土壤微生态的影响,解析土壤及微生物在连续多年枝条覆盖下的垂直变化特点,通过研究枝条覆盖(5年)土壤在0~10、10~20、20~30、30~40、40~50 cm的5种不同深度下的理化性质特点,并利用PacBio平台对细菌、真菌进行三代全长扩增子测序分析。其结果如下:(1)枝条覆盖有提高土壤肥力的作用,土壤浅层的有机质、碱解氮含量提升明显,增加50%左右,且随着土层深度的增加影响逐渐减弱;(2)枝条覆盖能提高微生物多样性,智能调节微生物丰度,提高微生物的均匀度;(3)土壤微生物在门水平上主要由变形菌门(Proteobacteria)和子囊菌门(Ascomycota)构成,占比34.00%以上,属水平上主要由朱氏杆菌属(Chujaibacter,占比11.00%以内)和假散囊菌属(Pseudeurotium,占比45.00%以内)组成,种水平上主要由Paraburkholderia_sartisoli(占比5.00%以内)和Pseudeurotium_sp(占比45.00%以内)组成,且在枝条覆盖下各土层相对丰度均低于地布覆盖,枝条覆盖对真菌丰度的影响程度大于细菌;(4)细菌功能主要为代谢(占比46.00%以上),真菌功能主要为腐生生物(占比29.00%~61.00%),均与主要门类呈极显著相关关系;枝条覆盖对细菌功能影响较小,对真菌功能影响较大。综上,枝条覆盖作为行内土壤管理一种新途径,对废旧枝条利用、提高土壤养分、平衡土壤微生物群落有重要作用,为后续枝条覆盖的研究与应用提供参考。

    Abstract:

    In order to clarify the impact of branch covering on soil microbiology in pear orchards and to analyze the vertical change characteristics of soil and microorganisms under continuous years of branch covering,the characteristics of physicochemical properties of branch covered(5 years)soil at five different depths of 0-10,10-20,20-30,30-40,and 40-50 cm were investigated and three generations of full-length amplicon sequencing of bacteria and fungi were analysed using the PacBio platform. The results were as follows:(1)Branch mulching significantly enhanced soil fertility,particularly in the shallow layer,where organic matter and alkali-hydrolyzable nitrogen content increased by approximately 50%. This effect diminished with increasing soil depth;(2)Branch covering improved microbial diversity,intelligently regulated microbial abundance,and improved microbial uniformity;(3)Soil microorganisms at the phylum level were mainly composed of Proteobacteria and Ascomycota,accounting for over 34.00%,and at the genus level mainly composed of Chujaibacter(<11.00%)and Pseudeurotium(<45.00%),and at the species level mainly composed of Paraburkholderia_sartisoli(<5.00%)and Pseudeurotium_sp(<45.00%),and the relative abundance of each soil layer under branch cover was lower than that of the control,branch covering had a greater effect on fungal abundance than bacterial;(4)Bacterial function was predominantly Metabolish(>46.00%)and fungal function was predominantly Saprotroph(29.00%-61.00%),both with highly significant relationships to major phyla. Branch covering had a lesser effect on bacterial function and a greater effect on fungal function. In conclusion,branch covering,as a new way of soil management in rows,had an important role in solving waste branches,improving soil nutrients and improving soil microbial community,which could provide reference for subsequent research and application of branch mulching.
