    摘要:

    为了探究不同有机物料还田对曲靖烟区土壤肥力及微生物群落的影响,依托旱地黄壤田间试验,基于高通量测序技术分析化肥与不同有机物料配施对土壤化学性质、微生物群落丰度和结构的影响。试验采用随机区组设计,共设5个处理,分别为单施化肥(CK)、化肥+腐熟牛粪15000 kg/hm2(T1)、化肥+腐熟牛粪15000 kg/hm2+芝麻饼肥300 kg/hm2(T2)、化肥+腐熟牛粪15000 kg/hm2+高碳基肥750 kg/hm2(T3)、化肥+腐熟牛粪15000 kg/hm2+生物有机肥750 kg/hm2(T4),其中T1处理为当地常规施用量。结果表明,(1)相较CK,添加有机物料的处理均能提高土壤pH和养分含量,其中T2处理改良效果较好,有效磷、腐植酸、富里酸、微生物生物量碳和微生物生物量氮含量分别显著提升68.49%、22.55%、18.84%、16.89%和21.17%。(2)T2处理特有的细菌和真菌扩增子变异序列数量最多,分别为1606和1059个,Alpha多样性分析同样表明T2处理土壤细菌和真菌群落的丰富度和多样性最高。(3)门水平上,各处理土壤细菌的优势菌门为变形菌门、放线菌门、绿弯菌门、芽单胞菌门和酸杆菌门,土壤真菌的优势菌门为子囊菌门、担子菌门和被孢霉门;T4处理具有最高的放线菌门、芽单胞菌门和子囊菌门相对丰度。(4)土壤微生物生物量氮、全碳、全氮含量和pH是影响土壤细菌群落丰度和结构的主要影响因素,胡敏酸和富里酸的比、碳氮比和富里酸是影响土壤真菌群落丰度和结构的主要影响因素,其中毛囊门的丰度与多数土壤环境因子显著相关。综上,在曲靖黄壤植烟区配施有机物料能够提升烤烟移栽后70 d根际土壤pH和养分含量,其中化肥、腐熟牛粪和芝麻饼肥配合施用在提升土壤养分、增强微生物丰富度与多样性方面效果最佳,可为曲靖烟区土壤保育提供科学施肥依据。

    Abstract:

    In order to explore the effects of different organic materials returning to the field on soil fertility and microbial community in Qujing tobacco area,based on the field experiment of dryland yellow soil,the changes of soil chemical properties,microbial community abundance and structure under the combined application of chemical fertilizer and different organic materials were analyzed by high-throughput sequencing technology. A randomized block design was used in the experiment,and a total of 5 treatments were set up,namely:single application of chemical fertilizer(CK),chemical fertilizer + decomposed cow dung 15000 kg/hm2(T1),chemical fertilizer +15000 kg/hm2 decomposed cow dung + 300 kg/hm2 sesame cake fertilizer(T2),chemical fertilizer + 15000 kg/hm2 decomposed cow dung + 750 kg/hm2 high carbon base fertilizer(T3),chemical fertilizer + 15000 kg/hm2 decomposed cow dung + 750 kg/hm2 biological organic fertilizer(T4),of which T1 treatment was the local conventional application amount. Results showed that:(1)Compared with CK,the addition of organic materials increased soil pH and nutrient content. Among them,T2 treatment had better improvement effects,and the contents of available phosphorus,humic acid,fulvic acid,microbial biomass carbon and microbial biomass nitrogen were significantly increased by 68.49%,22.55%,18.84%,16.89% and 21.17%,respectively.(2)The number of unique bacterial and fungal amplicon sequence variants in T2 treatment was the largest,which was 1606 and 1059,respectively. Alpha diversity analysis also showed that the richness and diversity of soil bacterial and fungal communities in T2 treatment were the highest.(3)At the phylum level,the dominant phylum of soil bacteria in each treatment was Proteobacteria,Actinobacteriota,Chloroflexi,Gemmatiomonadota and Acidobacteriota,and the dominant phylum of soil fungi was Ascomycota,Basidiomycota and Mortierellomycota. T4 treatment had the highest relative abundance of Actinobacteria,Gemmatimonadetes and Ascomycota.(4)Soil microbial biomass nitrogen,total carbon,total nitrogen content and pH were the main influencing factors affecting the abundance and structure of soil bacterial community. Humic acid and fulvic acid ratio,carbon and nitrogen ratio and fulvic acid were the main influencing factors affecting the abundance and structure of soil fungal community. The abundance of Mucoromycota was significantly correlated with most soil environmental factors. This study showed that the application of organic materials in Qujing yellow soil tobacco planting area could improve the pH and nutrient content of rhizosphere soil at 70 d after transplanting of flue-cured tobacco. Among them,the combined application of chemical fertilizer,decomposed cow dung and sesame cake fertilizer had the best effect in improving soil nutrients and enhancing microbial richness and diversity,which could provide scientific fertilization basis for soil conservation in Qujing tobacco planting area.
