    摘要:

    以温室大棚为主的设施农业长期种植易导致土壤板结、土壤酸化、土壤盐渍化等,进而导致土壤细菌群落失衡,严重制约设施蔬菜的可持续发展。利用高通量测序技术等研究了不同种植年限对设施菜地土壤细菌群落以及土壤化学性质的影响,并探究土壤化学因子与细菌群落变化的关联性。研究结果表明,随着种植年限的增加,土壤pH由5.85下降至5.11、全氮由1.29 g/kg增加至1.68 g/kg、碱解氮由120.22 mg/kg增加至222.40 mg/kg(P<0.05)、总盐度由1.97%增加至2.85%~3.27%。经过长达20年种植,细菌α多样性显著降低(P<0.05),经置换多元方差分析分析表明细菌群落结构发生显著变化(P<0.01)。在前10门、属中,变形菌门的相对丰度降至最低(20.69%),而厚壁菌门和绿弯菌门的相对丰度则分别增加了3.21%、22.14%,达到4.40%、30.40%;鞘氨醇单胞菌属和芽单孢菌属相对丰度分别下降3.66%、1.94%(P<0.05)。典型相关性分析表明pH、碱解氮是影响细菌群落结构的主要环境因子(P=0.011、P=0.036)。种植时间延长(>10年)导致土壤酸化及氮素积累,显著降低细菌群落多样性,改变细菌群落结构,进而引发土壤微生态失衡,制约设施蔬菜地的可持续利用。

    Abstract:

    Long-term cultivation in greenhouse facilities could lead to soil compaction,soil acidification,and soil salinization,which in turn caused imbalances in soil bacterial communities and severely restricted the sustainable development of facility vegetables.The high-throughput sequencing technology and other methods were utilized to investigate the effects of different planting years on soil bacterial communities and soil chemical properties in facility vegetable fields,and to explore the correlations between soil chemical factors and bacterial community changes.The results showed that as the planting years increased,soil pH decreased from 5.85 to 5.11,total nitrogen increased from 1.29 g/kg to 1.68 g/kg,available nitrogen increased from 120.22 mg/kg to 222.40 mg/kg(P<0.05),and total salinity increased from 1.97% to 2.85%-3.27%.After 20 years of planting,bacterial α-diversity significantly decreased(P<0.05),and PERMANOVA analysis indicated significant changes in bacterial community structure(P<0.01).Among the top 10 phyla and genera,the content of Proteobacteria decreased to the lowest level of 20.69%,while the contents of Firmicutes and Chloroflexi increased by 3.21% and 22.14%,respectively,reaching 4.40% and 30.40%.The relative abundances of Sphingomonas and Bacillus decreased by 3.66% and 1.94%,respectively(P<0.05).Canonical correlation analysis indicated that pH and available nitrogen were the main environmental factors affecting bacterial community structure(P=0.011,P=0.036).With the extension of planting time(>10 years),soil acidification and nitrogen accumulation occurred,significantly reducing the diversity of bacterial communities,altering the structure of bacterial communities,and subsequently causing an imbalance in soil microecology,which could restrict the sustainable utilization of facility vegetable land.
