    摘要:

    枯草芽孢杆菌(Bacillus subtilis)作为农业生产中广泛应用的益生菌,其高效增殖对微生物菌剂效能发挥至关重要。研究旨在明确微藻对枯草芽孢杆菌的促生潜力及最适作用浓度,筛选具有显著促生效果的微藻株并解析其作用方式。以极端环境分离的微藻与枯草芽孢杆菌为试验材料,通过初筛获得6株促生作用显著的藻种。并经18S rDNA、16S rDNA序列分析鉴定,6株藻种分别属于索罗金小球藻(Chlorella sorokiniana)、鱼腥藻(Anabaena sp.)、栅藻(Pseudospongiococcum sp.)。设置0%(对照CK)、20%、40%、60%、80%、100%浓度梯度,微藻的活藻培养液、上清液、浓缩液、破碎液处理,研究其对枯草芽孢杆菌生长的促进作用。结果表明,索罗金小球藻CCAXNU-001的浓缩液促生作用最为显著,随分泌物浓度升高,促生效应呈先增强后减弱的趋势,60%浓度时促生效果最佳,超过该阈值后作用减弱。微藻可溶性分泌物可显著促进枯草芽孢杆菌的增殖,其中浓缩液处理的促生效果优于活藻培养液、上清液和破碎液。研究明确了微藻对枯草芽孢杆菌的促生作用及浓度效应,为开发基于微藻-益生菌的高效微生物制剂提供了理论依据。

    Abstract:

    As a probiotic widely used in agricultural production,the efficient proliferation of Bacillus subtilis is crucial for the effectiveness of microbial inoculants.This study aimed to clarify the growth-promoting potential of microalgae on B. subtilis and the optimal concentration,screen microalgal strains with significant promoting effects,and analyze their action modes.Using microalgae isolated from extreme environments and B. subtilis as experimental materials,six microalgal strains with significant growth-promoting effects were obtained through primary screening.Identified by 18S rDNA,16S rDNA sequence analysis,the six strains belonged to Chlorella sorokiniana,Anabaena sp.,and Pseudospongiococcum sp.(Note: Pseudospongiococcum is a newly proposed genus).Concentration gradients of 0% (control CK),20%,40%,60%,80%,and 100% were set up,using live algal culture solutions,supernatants,concentrated solutions,and lysates of microalgae to study their promoting effects on B. subtilis growth.The results showed that the concentrated solution of Chlorella sorokiniana CCAXNU-001 exhibited the most pronounced growth-promoting effect.With the increase in secretory concentration,the promoting effect first increased and then decreased,reaching the optimal effect at 60%,after which the effect weakened.Soluble secretions of microalgae significantly promoted the proliferation of B. subtilis,with the concentrated solution treatment outperforming live algae,supernatants,and lysates.This study clarified the growth-promoting effect and concentration-dependent response of microalgae on B. subtilis,providing a theoretical basis for developing efficient microbial formulations based on microalgae-probiotic combinations.
