    摘要:

    探究利用微生物菌剂调控连作半夏土壤肥力及根际微生物群落来缓解半夏连作障碍,为消除半夏连作障碍提供参考。以连作1 年的半夏及半夏根际土壤为研究对象,设置对照(CK)、单施枯草芽孢杆菌剂(KC)、单施哈茨木霉菌剂(HC)、单施解淀粉芽孢杆菌剂(JD)、复合菌剂(ZH,配施枯草芽孢杆菌剂+ 哈茨木霉菌剂+ 解淀粉芽孢杆菌剂)5 种处理,对不同菌剂处理下半夏生长和土壤理化性质、酶活性及根际微生物群落进行研究。微生物菌剂显著促进了半夏的生长,ZH 处理的株幅、叶柄粗和产量效果最好,较CK 增加了11.62%、18.88% 和77.32%。4 种菌剂均能提高半夏根际土壤的理化性质和土壤酶活性,ZH 效果最好,土壤碱解氮、速效钾、有效磷和有机质含量比CK 提高了20.37%、18.62%、24.94% 和23.99%,土壤过氧化氢酶、碱性磷酸酶、脲酶和硝酸还原酶活性较CK 显著提高了3.38%、61.64%、28.00% 和57.74%。高通量测序分析表明,相对于单一菌剂处理和CK,ZH 处理增加了OTU 数、物种丰富度和多样性。在门水平上,ZH 处理提高了根际细菌中的假单胞菌门(Pseudomonadota)、酸杆菌门(Acidobacteriota)和放线菌门(Actinobacteriota)的相对丰度,降低拟杆菌门(Bacteroidota)和硝化螺旋菌门(Nitrospirota)相对丰度,降低了子囊菌门(Ascomycota)的相对丰度;从属水平上看,提高了鞘氨醇单胞菌属(Sphingomonas)和寡养杆菌属(Stenotrophobacter)的相对丰度,黄杆菌属(Lysobacter)、镰刀菌属(Fusarium)和丝核菌属(Rhizoctonia)的相对丰度显著降低。相关分析表明,碱解氮、速效钾与鞘氨醇单胞菌属呈显著正相关,有机质与浮霉菌门、接合菌门呈显著正相关,碱性磷酸酶活性与锥盖伞属(Conocybe)呈显著正相关,过氧化氢酶与小球腔菌属(Plectosphaerella)呈显著正相关,而镰刀菌属、裂孢壳属(Schizothecium)和黄杆菌属与多种土壤理化指标和酶活性呈显著负相关。微生物菌剂能促进半夏的生长发育,提高半夏土壤肥力,增加土壤微生物多样性及改良微生物群落结构,其中,以ZH 处理效果最佳,可作为缓解半夏连作障碍的有效菌剂。

    Abstract:

    The study aimed to explore the use of microbial agents to regulate the soil fertility and rhizosphere microbial community of continuous cropping Pinellia ternata,so as to alleviate the continuous cropping obstacle of P. ternata and provide reference for eliminating the continuous cropping obstacle of P. ternata. Taking P. ternata and its rhizosphere soil after continuous cropping for one year as the research object,five treatments were set up,including control(CK),single application of Bacillus subtilis agent(KC),single application of Trichoderma harzianum agent(HC),single application of Bacillus amyloliquefaciensagent(JD),and compound microbial agent(ZH,combined application of Bacillus subtilis agent + Trichoderma harzianum agent + Bacillus amyloliquefaciens agent). The growth,soil physical and chemical properties,enzyme activities and rhizosphere microbial community of P. ternata under different microbial agents were studied. Microbial agents significantly promoted the growth of P. ternata. The plant width,petiole diameter and yield of ZH treatment were the best,which increased by 11.62%,18.88% and 77.32%,respectively,compared with CK. The four microbial inoculants could improve the physical and chemical properties and soil enzyme activities of the rhizosphere soil of P. ternata. The effect of ZH treatment was the best. The contents of alkali-hydrolyzable nitrogen,available potassium,available phosphorus and organic matter in soil increased by 20.37%,18.62%,24.94% and 23.99%,respectively,compared with CK. The activities of soil catalase,alkaline phosphatase,urease and nitrate reductase were significantly increased by 3.38%,61.64%,28.00% and 57.74% compared with CK. High-throughput sequencing analysis showed that compared with single treatment and CK treatment,ZH treatment increased the number of OTU,species richness and diversity. At the phylum level,ZH treatment increased the relative abundance of Pseudomonadota,Acidobacteriota and Actinobacteriota in rhizosphere bacteria,decreased the relative abundance of Bacteroidota and Nitrospirota,and decreased the relative abundance of Ascomycota. At the genus level,the relative abundance of Sphingomonas and Stenotrophobacter was increased,and the relative abundance of Lysobacter,Fusarium and Rhizoctonia was significantly reduced. Correlation analysis showed that alkali-hydrolyzable nitrogen and available potassium were significantly positively correlated with Sphingomonas,organic matter was positively positively correlated with Planctomycetes and Zygomycota,alkaline phosphatase activity was positively correlated with Conocybe,catalase was positively correlated with Plectosphaerella,while Fusarium,Schizothecium and Lysobacter were negatively correlated with various soil physical and chemical indicators and enzyme activities. Microbial agents could promote the growth and development of P. ternata,improve soil fertility,increase soil microbial diversity and improve microbial community structure. Among them,ZH treatment had the best effect and could be used as an effective microbial agent to alleviate the continuous cropping obstacle of P. ternata.
