The effects of tobacco-buckwheat rotation on the structure and function of soil fungal communities in tobaccogrowing areas at different altitudes in Shimen county

    摘要:

    为了探究石门县不同海拔烟区烤烟- 荞麦轮作对土壤真菌群落的影响,在石门县选择3 个长期烤烟连作的地区,设置烤烟连作(YY)和烟荞轮作(YQ)2 个处理,采用高通量测序技术,对不同海拔烤烟连作和烟荞轮作土壤真菌群落多样性、结构和功能进行研究。结果表明,3 个地区轮作土壤的扩增子序列变体数量均有所升高,连作和轮作土壤真菌群落构成差异较大,轮作土壤真菌具有更高的丰富度和均匀度。所有土壤样品共检测到14 门、50 纲、130 目、343 科、756 属、1165 种,连作和轮作在土壤真菌门水平上均以子囊菌门为优势菌门,轮作降低了优势菌门的比例,增加了优势菌门的种类;轮作与连作相比,优势菌属均发生改变,轮作降低了镰刀菌属、小不整球壳属、链格孢属、角担菌属和Bisifusarium 的相对丰度,增加了葡萄孢属、Humicola、枝孢菌属、篮状菌属和Itersonilia 的相对丰度;主坐标分析表明,连作和轮作处理的土壤真菌群落组成差异显著。线性判别分析结果显示,各处理包含的差异物种在12 ~ 17 个之间。对土壤真菌进行功能分类,共预测得到8 种不同的营养类型,其中病理- 腐生- 共生营养型和腐生营养型在连作和轮作土壤均占比较高,占比分别为19.74% ~ 72.09% 和14.87% ~ 57.51%,轮作降低了病理营养型所占比例,增加了腐生营养型的比例,且新增了腐生- 病理- 共生营养型。综上,烟荞轮作提高了土壤真菌群落的丰富度和多样性,改变了土壤真菌的群落结构,对改善烤烟连作土壤微生态环境,缓解烤烟连作障碍具有积极作用。

    Abstract:

    To investigate the impact of tobacco-buckwheat rotation on the soil fungal communities in tobacco-growing areas at different altitudes in Shimen county,three regions with long-term continuous cropping of flue-cured tobacco in Shimen county were selected,and two treatments,continuous cropping of flue-cured tobacco(YY)and tobacco-buckwheat rotation(YQ),were set up. High-throughput sequencing technology was used to study the diversity,structure and function of the soil fungal communities of continuous cropping of flue-cured tobacco and tobacco-buckwheat rotation in different areas. The results showed that:the number of ASVs increased in rotation soils compared with the continuous cropping of all three regions.There were significant differences in the composition of soil fungal communities between continuous cropping and rotation.The rotation soil had higher richness and evenness of fungal communities. A total of 14 phyla,50 classes,130 orders,343 families,756 genera,and 1165 species were detected in all soil samples. At the phyla level,Ascomycota was the dominant phyla in both continuous cropping and rotation soil. Rotation reduced the proportion of dominant phylum and increased the number of dominant phylum species,compared with the continuous cropping. Compared with continuous cropping,rotation changed the dominant fungal genera. Rotation reduced the relative abundance of Fusarium,Plectosphaerella,Alternaria,Ceratobasidium,and Bisifusarium,while increasing the relative abundance of Botrytis,Humicola,Cladosporium,Talaromyces and Itersonilia. principal coordinate analysis showed that there were significant differences in the composition of soil fungal communities between continuous cropping and rotation treatments. Line discriminant analysis effect size analysis results showed that the number of differential species in each treatment ranged from 12 to 17. The soil fungi in the continuous cropping and rotation systems of the three regions were functionally classified,and a total of 8 different nutritional types were predicted. The Pathotroph-Saprotroph-Symbiotroph and Saprotroph nutritional types accounted for a relatively high proportion in both continuous cropping and rotation soils,with proportions ranging from 19.74% to 72.09% and from 14.87% to 57.51%,respectively. Rotation reduced the proportion of the Pathotroph nutritional type and increased the proportion of the Saprotroph nutritional type,and introduced the Saprotrophic-Pathotrophic-Symbiotrophic type. In conclusion,the tobacco-buckwheat rotation increased the richness and diversity of soil fungal communities,changed the structure of soil fungal communities,and had a positive effect on improving the soil micro-ecological environment of continuous cropping flue-cured tobacco and alleviating the continuous cropping obstacles of flue-cured tobacco.
