Effects of solid fermentation products of Trametes gibbosa SICAU-T41 on agronomy traits of flue-cured tobocco and rhizosphere soil microecology

    摘要:

    磷是土壤的主要元素之一,也是生命元素之一。溶磷菌对于土壤磷素的转化起着重要作用,有关溶磷真菌的研究主要集中在曲霉菌属、青霉菌属、镰刀菌属和木霉属等,但关于多孔菌目真菌土壤磷素的影响特征尚较少。本研究以浅囊状栓菌SICAU-T41 为研究对象,将其固体发酵基质进行了烤烟盆栽应用,并结合化学分析、荧光定量PCR 和高通量测序对其根际微生态效应进行了分析。研究结果显示,固体发酵基质显著促进烤烟生长发育。施加菌株SICAU-T41 固体发酵基质的处理(T4)烤烟株高、根长、最大叶长、最大叶宽、鲜重和茎粗分别提升133.87%、27.48%、75.17%、105.45%、299.04% 和42.04%;土壤有机质、碱解氮和有效磷含量增加。水溶性磷和碳酸氢钠提取性磷含量显著提升。T4 处理的菌株定殖量为3.38×107 copies/g,相较于液体菌剂处理的1.60×107 copies/g 提升了112%;土壤优势细菌门为变形菌门(Proteobacteria)、酸杆菌门(Acidiobacteriota)和放线菌门(Actinobacteriota)。真菌优势门为子囊菌门(Ascomycota)、被孢霉门(Mortierellomycota)和担子菌门(Basidiomycota);黏液杆菌属(Mucilaginibacter)、苍白杆菌属(Ochrobactrum)和鞘氨醇杆菌属(Sphingobacterium)等细菌和假丝酵母属(Candida)和丛赤壳菌属(Nectria)等真菌的相对丰度值显著提升。添加溶磷促生型菌株SICAU-T41 的固体发酵物可以提高烤烟的农艺性状,增加土壤养分含量,改变土壤磷素组分和微生物群落结构,具有重要应用价值。

    Abstract:

    Phosphorus is one of the major elements in soil and is indispensable for life activities.Phosphorus-solubilizing microorganisms play a very important role in the transformation of phosphorus in the soil.Currently,studies on phosphorussolubilizing fungi mainly focus on Aspergillus,Penicillium,Fusarium,Trichoderma,etc.,yet few studies focused on the phosphorus-solubilizing ability of Basidiomycetes.Therefore,this study used Trametes gibbosa SICAU-T41 as the research material,and applied the solid fermented product of strain SICAU-T41 on flue-cured tobacco pot cultivation experiments,chemical analysis,quantitative real-time polymerase chain reaction,and high-throughput sequencing were combined to analyze the rhizosphere microecosystem effects of strain on flue-cured tobacco.The results showed that the solid fermented product significantly promoted the growth and development of flue-cured tobacco.In the pot experiment,the treatmen(t T4) with the application of the solid fermented product of SICAU-T41 had significantly increased the plant height,root length,maximum leaf length,maximum leaf width,fresh weight,and stem diameter of flue-cured tobacco,which were 133.87%,27.48%,75.17%,105.45%,299.04%,and 42.04% higher than the control treatment,respectively.The contents of soil organic matter,alkali-hydrolyzed nitrogen,available phosphorus,water-soluble phosphorus and phosphorus extracted by sodium bicarbonate increased significantly( P<0.05).The qPCR quantitative results showed that the colonization amount of the strain in the T4 treatment reached 3.38×107 copies/g,which was increased by 112% compared with that of the liquid microbial inoculum treatmen(t 1.60×107 copies/g).The dominant bacterial phyla in the soil were Proteobacteria,Acidobacteriota,and Actinobacteriota,while the dominant fungal phyla were Ascomycota,Mortierellomycota,and Basidiomycota.The relative abundances of Mucilaginibacter,Ochrobactrum,and Sphingobacterium,and Candida,and Nectria were significantly increased.The solid-state fermentation product of strain SICAU-T41 enhanced the fluecured tobacco agrcnomic performance,increased the soil nutrients contents,and varied the soil microbial communities,and showed prospective application value.
